Jazz Pharmaceuticals has updated that its lung cancer drug, Zepzelca, failed to meet the goal of improving overall survival in a late-stage study. The company has shared the trial results with U.S. Food and Drug Administration and will discuss the nest steps for Zepzelca as second-line indication. The Phase 3 treatment fell short on overall survival in the second-line treatment of relapsed metastatic small cell lung cancer.
Lagoon Trial Evaluates Zepzelca in SCLC Patients
This late-stage confirmatory trial tested Zepzelca as a single therapy as well as in combination with chemotherapy. Lagoon trial, a Phase III study sponsored by PharmaMar was conducted across more than 200 sites globally. The Lagoon trail enrolled 724 patients with second-line small cell lung cancer whose disease had progressed prior platinum-based chemotherapy.
Trial Results and Survival Outcomes
In this study, the lung cancer patients were treated with Zepzelca alone lived a median of 8.7 months and those on combination therapy lived 10.9 months.
Factors That May Have Influenced the Results
One of the complicating factors of the results was that this Lagoon trail was based on broader patient population in comparison to Phase II pivotal study.

How the IMforte Trial Differs
The full U.S. approval of Zepzelca in 2025 is based on Phase 3 IMforte tria, which evaluated this drug along with atezolizumab as first-line maintenance treatment for extensive-stage SCLC patients. These Lagoon results are distinct from IMforte trial.
